A Virtual Drive is a shortcut to any folder that may be located deep in the file system mapped to a drive letter, just like in Windows. Once created, these drives appear as regular drives in any File Explorer or Manager on your Nokia S60 3rd Edition mobile device. You can even map system folders if you have the appropriate hacks applied. Moreover, you can also see these drives via Nokia PC browser on your computer when connected via USB.
Here are two different applications that allow you to create and add virtual drives on Nokia S60:

Map Drives: Map Drives from Symbian-Freak can be downloaded from here. After installation, in your E drive, you will find a file called MapDrives.txt.
Edit it with the following syntax:
V=E:\Foldername\
Few things to note are:
+ The drive unit must be a Capital letter
+ The source path must end in BackSlash
Griffon: This is another freeware application to map drives on Nokia S60. You just need to install it and restart the mobile device. You can download this program from here.
